What is Kisspeptin-10?

Kisspeptin-10 (KP-10) is the C-terminal decapeptide fragment of kisspeptin (amino acids 112-121 of the parent protein), a hypothalamic neuropeptide encoded by the KISS1 gene that is a key upstream regulator of reproductive hormone release.

It is a research tool, not an approved drug: human evidence consists mainly of controlled physiology studies (notably by Dhillo and colleagues) examining its effects on GnRH-driven LH/FSH secretion.

There is no approved therapeutic product; use is confined to research settings.

How does Kisspeptin-10 work?

Kisspeptin-10 binds the KISS1R (GPR54) receptor on hypothalamic GnRH neurons, stimulating pulsatile GnRH release, which in turn drives luteinizing hormone (LH) and follicle-stimulating hormone (FSH) secretion from the pituitary.

What does the research say about Kisspeptin-10?

  • In healthy men, a single intravenous bolus of kisspeptin-10 caused a rapid, dose-dependent rise in serum LH at doses as low as 0.3 nmol/kg, and continuous infusion increased LH pulse frequency and testosterone. [1]
  • Kisspeptin-10's effect is sexually dimorphic: it stimulated LH/FSH in men and in women during the preovulatory phase, but produced no significant gonadotropin rise in women during the follicular phase even at high doses. [2]
  • The LH response to kisspeptin-10 is modulated by sex-steroid feedback, being enhanced in sex-steroid-deficient (e.g., postmenopausal) states, illustrating its role at the top of the reproductive axis. [3]

Side effects & safety profile

In the short controlled human physiology studies conducted, kisspeptin-10 was generally well tolerated with no serious adverse events reported, but these studies were small, acute (single bolus or short infusion), and conducted under medical supervision.

There is no long-term human safety data, no approved product, and no established chronic dosing, so any use outside research is unstudied.

Because it acts on the reproductive hormone axis, effects on fertility hormones are expected and unmonitored self-use carries unknown endocrine risks.
